ABSTRACT
The budding yeast Saccharomyces cerevisiae has a closed mitosis in which the mitotic spindle and cytoplasmic microtubules (MTs), both of generate forces order to faithfully segregate chromosomes, remain separated by nuclear envelope throughout cell cycle. Kar3, kinesin-14, distinct functions on MTs each compartment. Here, we show that two proteins, Cik1 Vik1, form heterodimers with regulate its localization function within along cycle-dependent manner. Using MT dynamics reconstitution assay lysates from cycle-synchronized cells, found Kar3Vik1 induces catastrophes S phase metaphase limits polymerization G1 anaphase. In contrast, Kar3Cik1 promotes pauses G1, while increasing Adapting this track motor protein motility, observed is necessary for Kar3's ability plus-ends metaphase, but, surprisingly, not during These experiments demonstrate how binding partners Kar3 modulate diverse spatially temporally.
