ABSTRACT
Renewal of stem cells differs from cancer cell growth in self-controlled division. The mir-302 microRNA (miRNA) family (mir-302s) is expressed most abundantly slow-growing human embryonic (ES) cells, and quickly decreases after differentiation proliferation. Therefore, mir-302s was investigated as one the key factors essential for maintenance ES renewal pluripotency this study. Pol-II-based intronic miRNA expression system used to transgenically transfect into several lines. mir-302-transfected namely, miRNA-induced pluripotent (mirPS) not only many markers, such Oct3/4, SSEA-3, SSEA-4 ,Sox2, Nanog, but also had a highly demethylated genome similar reprogrammed zygotic genome. Microarray analyses further revealed that genome-wide gene patterns between mirPS H1 H9 shared over 86% similarity. Using molecular guidance vitro, these could differentiate distinct tissue types, neuron-, chondrocyte-, fibroblast-, spermatogonia-like primordial cells. Based on findings, we conclude function reprogram an ES-like state maintain under feeder-free cultural condition, which may offer great opportunity therapeutic intervention.
