ABSTRACT
BACKGROUND Carbon ion radiotherapy has been shown to be more effective in cancer than photon irradiation. Influence of carbon radiation on microenvironment is very important for the outcomes radiotherapy. Tumor-infiltrating dendritic cells (DCs) play critical roles antigen processing and antitumor immunity. However, there scant literature covering effects DCs. In this study, we aimed uncover impact irradiation bone marrow derived MATERIAL AND METHODS Bone were co-cultured with GM-CSF IL-4 seven days, population DCs was confirmed flow cytometry. We used an Annexin V PI staining method detect cell apoptosis. Endocytosis assay determined by using a cytometry method. migration capacity tested Transwell also ELISA western blotting examine cytokines protein expression, respectively. RESULTS Our data showed that induced apoptosis both immature mature After irradiation, endocytosis impaired. Interestingly, triggered burst IFN-g IL-12 LPS or CpG treated DCs, which provide novel insights into combination immunotherapy Finally, found suppression p38 dependent. CONCLUSIONS present study demonstrated impaired function mainly through signaling pathway. anti-tumor secretion. This work provides information new immune adjuvant
