ABSTRACT
BACKGROUND:Loss of the epithelial barrier is characterized by a reduction in E-cadherin expression and hallmark asthma. Qi-xian decoction (QXT) Chinese medicinal formula that has been used to effectively treat This study aimed investigate effect QXT on human lung 16HBE cells ovalbumin-challenged mice explore underlying molecular mechanism. MATERIAL AND METHODS:Ovalbumin (OVA)-induced were as model Real-time PCR Western blotting utilized examine mRNA protein levels. Lung tissue reactive oxygen species (ROS) levels evaluated using dichloro-dihydro-fluorescein diacetate (DCFH-DA). Serum superoxide dismutase (SOD) total antioxidant capacity (TAOC) measured via enzyme-linked immunosorbent assay (ELISA)-based analyses. or hydrogen peroxide (H₂O₂) vitro. RESULTS:We found treatment increased decreased extracellular-signal-regulated kinase (ERK) phosphorylation tissues OVA-challenged mice. also downregulated ROS serum SOD TAOC In vitro studies demonstrated generation induced H₂O₂ resulted cells, which was attenuated inhibition ERK signaling. Moreover, H₂O₂-induced downregulation expression, generation, activation restored with water ethanol extract. CONCLUSIONS:These data demonstrate one mechanism protects against asthma restore vivo inhibiting ROS-mediated activation.
