ABSTRACT
One of the reasons to causing blood coagulation in tissue xenografted organs was known incompatibility and anti-coagulation regulatory system between TG pigs primates. Thus, overexpression human CD73 (hCD73) pig endothelial cells is considered as a method reduce coagulopathy after pig-to-non-human-primate xenotransplantation. This study performed produce breed transgenic expressing hCD73 for studies immune rejection responses could provide successful application The were constructed an expression vector under control porcine Icam2 promoter (pIcam2-hCD73) established donor cell lines hCD73. numbers transferred reconstructed embryos 127 ± 18.9. pregnancy delivery rate surrogates 8/18 (44%) 3/18 (16%). total number delivered cloned 10 (2 alive, 7 mummy, 1 died birth). Among them, three live hCD73-pigs successfully by Caesarean section, but one dead birth. two had normal reproductive ability. They mated with wild type (WT) MGH (Massachusetts General Hospital) female sows produced totally 16 piglets. 5 piglets identified pigs. In conclusion, we generated their litters natural mating. It can be possible use mate production multiple such α-1,3-galactosyltransferase knock-out /hCD46
