ABSTRACT
Numerous chemicals are capable of disrupting androgen production, but the possibility that they might act together to produce effects greater than those most effective component in mixture has not been studied directly human tissues. Suppression synthesis fetal life associated with testis maldescent, malformations genitalia at birth, and poor semen quality later life. Our aim was investigate whether can disrupt production explants evaluate importance when characterizing hazard individual chemicals. We used an organotypic culture system testes called FEtal Gonad Assay (FEGA) tissue obtained 10 12 gestational wk (GW 10-12), screen 27 individually for their possible anti-androgenic effect. Based on results screen, we selected 11 compounds tested them as mixtures. We evaluated mixtures composed four eight antiandrogens contained pharmaceuticals ketoconazole theophylline several previously untested chemicals, such pesticides imazalil propiconazole. Mixtures suppress testosterone testicular extent seen chemicals. This revealed itself a shift towards lower doses dose-response curves became more pronounced number components increased from eight. Our FEGA provide foundations predictive risk assessment approach exposures life. https://doi.org/10.1289/EHP1014.
