ABSTRACT
INTRODUCTION: Melatonin is a promising therapeutic agent due to its multiple beneficial effects, wide availability and relatively high safety. As melatonin acts as chronobiotic agent, adequate production by the pineal gland allows for adaptation environmental changes, while disturbances in secretion are associated with health disorders. The renoprotective effect of exogenous was established on different experimental models acute kidney injury (AKI), influence altered activity efficacy treatment has not been investigated.
OBJECTIVES: aim this research study potential conditions aminoglycoside-induced AKI against background hypo- hyperfunction.
METHODS: Nonlinear mature white rats (n=40) were randomly divided into 5 groups. Animals from I (Control), II (AKI) group kept under natural light regimen. Pineal hypofunction simulated III maintenance constant at 500 lux (24.00 : 0.00 darkness) 7 days. hyperfunction IV darkness (0.00 24.00 darkness). Toxic (II-IV groups) induced daily administration gentamicin dose 80 mg/kg 6 III-IV groups injected mg/kg. 24 h after last injection biochemical histological examination performed. For statistical analysis SPSS 17.0 software used.
RESULTS: Nephrotoxicity caused significant (p<0.05) functional changes structural alterations rat kidneys. Treatment gentamicin-induced significantly limited degree damage renal tissue prevented critical reduction function, confirming protective melatonin. At same time, differences between indices allow us state, that endogenous deficiency less effective comparison CONCLUSION: ameliorates limitation histopathological preservation function. Pre-existing decreases resistance kidneys damaging action toxin lessens Alternatively, increased production, co-treatment more effectively protects prevents development failure.
