Zoledronic Acid and Renal Toxicity: Data from French Adverse Effect Reporting Database

ABSTRACT
Zoledronic acid-associated renal impairment leading to failure has been recently reported in a cohort of US patients. However, the presence such toxicity other populations not yet determined.To analyze French cases zoledronic nephrotoxicity.We evaluated available with acute deterioration function associated acid therapy drawn from Adverse Event Reporting System database until July 1, 2004.We identified 4 men and 3 women between ages 52 70 years, multiple myeloma or different types metastatic cancer, who had experienced during therapy. Four patients developed de novo failure, while preexisting chronic failure. Renal occurred after various durations (1-120 days). Three completely recovered one partially their previous discontinuation acid, but was fatal outcome 2 cases; case unknown. Our data confirm previously risk factors for nephrotoxicity as advanced myeloma, diabetes, hypertension, concomitant use nephrotoxic drugs.These emphasize need regular monitoring treatment, particular attention impaired function.
