ABSTRACT
Gata-3 has been shown to specifically alter its expression patterns in different types of cancers. Recent evidence suggests that an interference exists the TGF-beta signaling pathway. To determine role pancreatic cancer, cancer samples were analyzed comparison normal tissues. Furthermore, four cell lines with alterations pathway studied. evaluate if a potential relationship exists, we correlated mRNA levels TGF-betas, receptors, and Smad-3. Finally, influence on vitro. All demonstrated marked overexpression protein. Immunohistochemical staining revealed strong persistent cytoplasmic immunoreactivity cells. In electrophoretic mobility shift assay, disturbed nuclear translocation was confirmed. The showed significant correlation responsive downregulation upon exposure, whereas TGF-beta-unresponsive lines, persisted at high levels. specific upregulation impaired cooperative action pathway, suggesting plays central human cancer.
