ABSTRACT
15-Lipoxygenase-1 (15-LOX-1) oxidizes polyunsaturated fatty acids to a rich spectrum of biologically active metabolites and is implicated in physiological membrane remodelling, inflammation apoptosis. Its deregulation involved the pathogenesis diverse cancer immune diseases. Recent experimental evidence reveals that dynamic histone methylation/demethylation mediated by methyltransferases demethylases plays critical role regulation chromatin remodelling gene expression. In present study, we compared 3 lysine 4 (H3-K4) methylation status 15-LOX-1 promoter region two Hodgkin lymphoma (HL) cell lines L1236 L428 with abundant undetectable expression, respectively. We identified potential H3-K4 positive transcription. Furthermore, found methyltransferase SMYD3 inhibition reduced expression decreasing activity cells. knock down these cells abolished di−/trimethylation H3-K4, attenuated occupancy transactivator STAT6, led diminished H3 acetylation at promoter. contrast, SMCX, JmjC-domain-containing tri-demethylase, upregulated through induction trimethylation, STAT6 recruitment addition, observed strong prostate line LNCaP its decreased Taken together, our data suggest important
