ABSTRACT
Quantitative real-time reverse-transcriptase PCR (qRT-PCR) is an important technique for analyzing differences in gene expression due to its sensitivity, accuracy and specificity. However, the stability of reference genes necessary ensure accurate qRT-PCR assessment interest. Perennial ryegrass (Lolium perenne L.) forage turf grass species temperate regions, but under various stresses has not been well-studied.In this study, 11 candidate were evaluated use as controls quantify perennial drought, high salinity, heat, waterlogging, ABA (abscisic acid) treatments. Four approaches--Delta CT, geNorm, BestKeeper Normfinder used determine these genes. The results are consistent with idea that best depend on stress treatment investigation. Eukaryotic initiation factor 4 alpha (eIF4A), Transcription elongation 1 (TEF1) Tat binding protein-1 (TBP-1) three most stably expressed drought also studying salt stress. eIF4A, TBP-1, Ubiquitin-conjugating enzyme (E2) suitable study heat stress, while TEF1, E2 effects ABA. Finally, Ubiquitin (UBQ), eIF4A waterlogging treatments.These will be helpful choosing studies related abiotic ryegrass. enable better normalization quantification transcript levels such studies.
