Performance of Simplexa Dengue Molecular Assay Compared to Conventional and SYBR Green RT-PCR for Detection of Dengue Infection in Indonesia

ABSTRACT
Diagnostic tests based on detection of dengue virus (DENV) genome are available with varying sensitivities and specificities. The Simplexa Dengue assay (Focus Diagnostics) is a newly developed real-time RT-PCR method designed to detect serotype DENV simultaneously. To assess the performance assay, we performed comparison conventional SYBR Green patients sera isolated from eight cities across Indonesia, endemic country. A total 184 that were confirmed using NS1 and/or IgM IgG ELISA examined. Using RT-PCR, detected in 53 (28.8%) 81 (44.0%) out sera, respectively. When was employed, rate increased 76.6% (141 samples). tested 40 by isolation as gold standard, yielded 95% sensitivity while reached 97.5% 100%, specificities all methods 100% when 43 non-dengue illness 20 healthy human samples. Altogether, our data showed higher compared field/surveillance setting. In conclusion, offers rapid accurate typing infection suitable for both routine diagnostic surveillance.
