Effects of Endocrine Disruptor Compounds, Alone or in Combination, on Human Macrophage-Like THP-1 Cell Response

ABSTRACT
The aim of the present study was to evaluate immunological effects on human macrophages four endocrine disruptor compounds (EDCs) using differentiated THP-1 cell line as a model. We studied first these EDCs, including Bisphenol A (BPA), di-ethylhexyl-phthalate (DEHP), dibutyl phthalate (DBP) and 4-tert-octylphenol (4-OP), either alone or in combination, cytokine secretion, phagocytosis. then determined whether not were mediated by estrogen receptors via MAPK pathways. It found that all EDCs reduced strongly phagocytosis cells several disturbed also TNF-α, IL-1 β IL-8 secretions. Furthermore, relative control treatment, decreased ERK 1/2 phosphorylation always associated with treatments—either certain combinations (at 0.1 μM for each condition). Lastly, treatments an receptor antagonist suppressed negative observed treated BPA, DEHP, 4-OP combined treatment BPA we suggested receptor-dependent pathway is involved mediating immune system. Altogether, results advocate can disturb response at very low concentrations.
