ABSTRACT
Background Parasitic helminths need to suppress the host immune system establish chronic infections. Paradoxically, immunosuppression induced by worm also benefits limiting excessive inflammation and tissue damage, which remains major cause leading serious morbidity mortality. Regulatory T cells (Tregs) are key regulators of this mutualism. The successive rise in Tregs during schistosome infection plays a critical role immunoregulation. We others previously showed that Schistosoma japonicum (S. japonicum) egg antigens (SEA) induce both vitro vivo. In addition, we identified SjHSP60 derived from SEA significantly induces vivo vitro. However, contribution Treg induction related mechanisms have not yet been identified. Methodology/Principal Findings study, S. stress protein HSP60 (SjHSP60) was constitutively extensively expressed eggs japonicum. specially without inducing other CD4+ sub-populations including Th1, Th2 Th17 cells. Furthermore, SjHSP60-depleted almost lost ability displayed significant impaired Finally, our study illustrated SjHSP60-mediated through conversion CD4+CD25- into CD4+CD25+Foxp3+ expansion preexisting TLR4-dependent manner. Conclusions/Significance Collectively, findings identify as parasitic contributor antigens, only contributes better understanding mechanism immunoregulation helminth infection, but suggests its potential therapeutic target for control immunopathology, allergic autoimmune diseases.
