ABSTRACT
Previous animal studies have shown that long term rat treadmill running induces over-use tendinopathy, which manifests as proteoglycan accumulation and chondrocytes-like cells within the affected tendons. Creating this model of tendinopathy by is however time-consuming, costly may vary among animals. In study, we used a new approach to develop an using kartogenin (KGN), bio-compound can stimulate endogenous stem/progenitor differentiate into chondrocytes. KGN-beads were fabricated implanted Achilles Five weeks after implantation, chondrocytes found at KGN site. Vascularity well disorganization in collagen fibers also present same site along with increased expression chondrocyte specific marker, type II (Col. II). vitro confirmed was released continuously from KGN-alginate vivo beads induced chondrogenic differentiation tendon (TSCs) suggesting chondrogenesis KGN-bead implantation tendons likely due aberrant TSCs Taken together, our results showed be create which, least part, reproduces features created running. This mechanistic (stem cell differentiation), highly reproducible precise creating localized tendinopathic lesions. It expected will useful evaluate effects various topical treatments such NSAIDs platelet-rich plasma (PRP) for treatment tendinopathy.
