Functional MRI for characterization of renal perfusion impairment and edema formation due to acute kidney injury in different mouse strains

ABSTRACT
Purpose The purpose was to characterize acute kidney injury (AKI) in C57BL/6 (B6)- and 129/Sv (Sv)-mice by noninvasive measurement of renal perfusion tissue edema using functional MRI. Methods Different severities AKI were induced B6- Sv-mice ischemia reperfusion (IRI). Unilateral clamping the pedicle for 35 min (moderate AKI) or 45 (severe done. MRI (7-Tesla) performed 1, 7 28 days after surgery a flow alternating inversion recovery (FAIR) arterial spin labeling (ASL) sequence. Maps T1-relaxation time calculated. Relative MRI-parameters IRI compared contralateral not-clipped between mouse strains unpaired t-tests. In addition, fibrosis assessed Masson Trichrome collagen IV staining. Results After moderate relative impairment significantly higher than at d7 (55±7% vs. 82±8%, p<0.05) d28 (76±7% 102±3%, p<0.01). T1-values increased early phase both strains. T1-increase more severe prolonged times strains, measured cortex outer stripe medulla. Kidney volume loss (compared kidney) occurred already but proceeded markedly towards 4 weeks AKI. Early predictive later loss. progression chronic disease (CKD) model similar as revealed histology. Conclusion Quantification allows characterization strain differences upon Renal stronger Sv-animals following Prolonged associated with formation CKD within observation.
