ABSTRACT
Eukaryotic protein phosphatase 4 (PP4) is a PP2A-type that part of conserved complex with regulatory factors PSY2 and PP4R2. Various lines Arabidopsis thaliana mutated PP4 subunit genes were constructed to study the so far completely unknown functions in plants. Mutants knocked out putative functional homolog LIKE (PSY2L) gene dwarf bushy, while plants PP4R2 (PP4R2L) looked very similar WT. The psy2l seedlings had short roots disorganized morphology impaired meristem. Seedling growth was sensitive genotoxin cisplatin. Global transcript analysis (RNA-seq) rosette leaves revealed several groups genes, shared between both types tissues, strongly influenced by PSY2L. Receptor kinases, CRINKLY3 WAG1, important for development, down-regulated 3-7 times. EUKARYOTIC ELONGATION FACTOR5A1 4-6 fold. Analysis hormone indicated abscisic acid levels high, auxin, cytokinin gibberellic low psy2l. Expression specific transcription involved regulation anthocyanin synthesis elevated, e.g. master regulator PAP1, intriguingly TT8, which otherwise mainly expressed seeds. mutants accumulated anthocyanins under conditions where WT did not, pointing PSY2L as possible upstream negative PAP1 TT8. sugar-phosphate transporter GPT2, cellular sugar phosphate homeostasis, enhanced 7-8 Several DNA damage response including cell cycle inhibitor WEE1, up-regulated activation repair signaling combination phenotypic traits showing aberrant root meristem sensitivity genotoxic cisplatin, substantiate involvement maintenance genome integrity.
