ABSTRACT
A new member of the genus Deltapartitivirus was identified containing three dsRNAs with an estimated size 1.71, 1.49 and 1.43 kb. The were extracted from symptomless pigeonpea [Cajanus cajan (L.) Millspaugh] plants cv. Erra Kandulu. This virus 4.64 kb genome tentatively named Arhar cryptic virus-1 (ArCV-1). genomic RNAs amplified characterized by sequence independent single primer amplification. shared a highly conserved 16 nt 5' non-coding region (5'-GATAATGATCCAAGGA-3'). largest dsRNA (dsRNA-1) as viral RNA dependent polymerase (replicase), predicted to encode putative 55.34 kDa protein (P1). two other smaller (dsRNA-2 dsRNA-3) for capsid proteins 38.50kDa (P2) 38.51kDa (P3), respectively. Phylogenetic analysis indicated that ArCV-1 formed clade together Fragaria chiloensis virus, Rosa multiflora Rose virus-1, indicating could be Deltapartitivirus. 3Dpol structure revealed several interesting features. in its full-length shares structural similarities members family Caliciviridaeand Picornaviridae. In addition, fourth molecule (dsRNA-2A), not related genome, found same plant tissue. dsRNA-2A (1.6 kb) encodes (P4), 44.5 kDa. P4 similarity coat genes viruses, particular bipartite viruses including Raphanus sativus virus-3. is first report occurrence plants.
