ABSTRACT
Asthma is a multifactorial pathology influenced by environmental and genetic factors. Glucocorticoid treatment decreases symptoms regulating genes involved in the inflammatory process through binding to specific DNA sequences. Polymorphisms located promoter region of Prostaglandin D Receptor (PTGDR) gene have been related asthma. We aimed analyze effect PTGDR haplotypes on expression response corticosteroid therapy. A549 lung epithelial cells were transfected with vectors carrying four different (CTCT, CCCC, CCCT TCCT), treated dexamethasone. Different approaches study activity (Dual Luciferase Reporter System), levels (qPCR) cytokine secretion (Multiplexed Bead-based Flow Cytometric) used. In addition, silico analysis was also performed. Cells TCCT haplotype showed lowest (p-value<0.05) mRNA basal conditions. After dexamethasone treatment, wild-type variant CTCT highest response, those luciferase assays. transcription factor patterns identified silico. Moreover, differences found among haplotypes. influence expression, as well secretory pattern. Furthermore, an association between these positions corticoid observed.
