ABSTRACT
We have developed our original tissue engineering technology “cell sheet engineering” utilizing temperature-responsive culture dishes. The cells are confluently grown on a dish and can be harvested as cell by lowering temperature without enzymatic digestion. Cell sheets high-cell-density tissues similar to actual living tissues, maintaining their structure function. Based this engineering”, we trying create functional cardiac from human induced pluripotent stem cells, for regenerative therapy in vitro drug testing. Toward purpose, it is necessary evaluate the contractility of engineered sheets. Therefore, present study, contractile force measurement system evaluated iPSC-derived sheet-tissues. By attaching fibrin gel sheets, created dynamically beating They were mounted was measured stably clearly. absolute values around 1 mN, mean value per cross-sectional area 3.3 mN/mm2. These equivalent or larger than many previously reported values, indicating functionality also confirmed that both rate significantly increased administration adrenaline, which physiologically relevant responses tissues. In conclusion, study valuable evaluation sheet-tissues, testing well.
