ABSTRACT
Background During normal pregnancy, placental oxidative stress (OS) is present during all three trimesters and necessary to obtain cell function. However, if OS reaches a certain level, pregnancy complications might arise. In preeclampsia (PE), dangerous specific hypertensive disorder, induced in the ischemic placenta causes systemic inflammatory response activates maternal endothelial cells. this study, we aimed quantify superoxide concentrations (as measure of OS) using electron paramagnetic resonance (EPR) correlate them markers inflammation, iron status vascular Methods Fifty-nine women with healthy (HP), 10 non-pregnant controls (NP) 28 PE patients (32±3.3weeks) were included. HP, blood samples for superoxide, neutrophil lymphocyte ratio (NLR), mean platelet volume (MPV) taken at 10, 25 39 weeks. Vascular measurements arterial stiffness (carotid-femoral pulse wave velocity (CF-PWV), augmentation index (AIx), Pressure (AP)) microvascular function (reactive hyperemia (RHI)) performed 35 PE, diagnosis. CMH (1-hydroxy-3-methoxycarbonyl-2,2,5,5-tetramethylpyrrolidine) was used as spin probe EPR, since formed CM radical corresponds amount superoxide. Results Superoxide concentration remains stable (p = 0.92), but significantly higher compared NP (p<0.0001). At weeks, there significant positive correlation between ferritin concentration. 0.04) inflammation are much HP (all p<0.001). p<0.04), while correlated 0.03). Conclusions an increased environment, which If levels increase, stiffness. pregnancies, result deterioration Together, these findings support hypothesis that directly linked measurement combination tests be prediction PE.
