ABSTRACT
Background Recent evidence suggests that antibiotic-induced changes in the composition of intestinal microflora, as well systemic immunoendocrine effects result from them, can modulate myocardial tolerance to ischemia-reperfusion injury. The aim this study was investigate tetracycline (TTC) on infarct size isolated hearts obtained obese rats with chemically-induced colitis (CIC). association between TTC-induced and microbiome plasma levels cytokines short-chain fatty acids (SCFAs) also studied. Methods Obesity induced Wistar by feeding them a high-fat, high-carbohydrate diet for five weeks. A single rectal administration 3% acetic acid (2 mL) resulted CIC. Healthy CIC received TTC (15 mg daily 3 days) via gavage. were euthanized, after which heart perfusion simulated global ischemia reperfusion performed. Infarct determined histochemically. Lipopolysaccharide (LPS) cytokine measured enzyme-linked immunosorbent assay, whereas SCFA gas chromatography/mass spectrometry. analyzed using reverse transcription polymerase chain reaction. Results treatment significant limitation (50 ± 7 vs. 62 4% control mice, p < 0.05) intact animals. However, not different Furthermore, significantly larger TTC-treated than it animals (77 5%, 0.05). concentrations proinflammatory LPS serum elevated Compared rats, both obesity had lower counts Lactobacillus Bifidobacterium spp. but higher Escherichia coli. associated specific levels. Conclusions reduced healthy rats. effect reversed Additionally, gut microbiota LPS.
