ABSTRACT
Multiplexed bead-based assays that use Luminex ® xMAP technology have become popular for measuring antibodies against proteins of interest in many fields, including malaria and more recently SARS-CoV-2/COVID-19. There are currently two formats widely used: non-magnetic beads or magnetic beads. Data lacking regarding the comparability results obtained using these types beads, run on different instruments. Whilst can only be flow-based instruments (such as 100/200™ Bio-Plex 200), both newer MAGPIX In this study we utilized a panel purified recombinant Plasmodium vivax samples from malaria-endemic areas to measure P . -specific IgG responses combinations We directly compared: i) versus 200, ii) 200 iii) also performed an external comparison our optimized assay. observed antibody responses, measured proteins, were moderately-strongly correlated all three comparisons (pearson r>0.5 18/19 proteins), however higher amounts protein required coupling Our indicated generated laboratories same coupled highly comparable r>0.7), particularly if reference standard curve is used.
