Downregulation of SFRP2 facilitates cancer stemness and radioresistance of glioma cells via activating Wnt/β-catenin signaling

ABSTRACT
Secreted frizzled-related protein 2 (SFRP2) is a glycoprotein with frizzled-like cysteine-rich domain that binds Wnt ligands or frizzled receptors to regulate signaling. SFRP2 frequently hypermethylated in glioma patients, and analysis of TCGA data indicates one the most downregulated genes radiotherapy treated patients. In present study, we aimed explore potential function tumorigenesis radioresistance glioma. The RNA sequencing samples were downloaded analyzed. expression 166 patients was evaluated by qRT-PCR. functions loss-of-function assays gain-of-function cell lines. We found radiotherapy-treated low correlated advanced tumor stage poor prognosis. CRISP/Cas9-meidated knockdown promoted soft agar colony formation, cancer stemness cells, while enforced exhibited opposite effects. Moreover, Wnt/β-catenin signaling activated lines, overexpression inhibited activation. Besides, pharmacological inhibition XAV-939 abrogated effects on cells. Our for first time demonstrated role suggested might be strategy increasing radiosensitivity
