ABSTRACT
Objective To identify the most effective fraction of Nanocnide lobata in treatment burn and scald injuries determine its bioactive constituents. Methods Chemical identification methods were used to analyze solutions extracted from using petroleum ether, ethyl acetate, n-butanol a variety color reactions. The chemical constituents extracts identified by ultra-performance liquid chromatography (UPLC)–mass spectrometry (MS). A total 60 female mice randomly divided into following 6 groups: ether extract-treated group; acetate model control positive drug group. burn/scald was established Stevenson’s method. At 24 hours after modeling, 0.1 g corresponding ointment evenly applied wound each Mice group did not undergo treatment, while those received Vaseline. Wound characteristics, including color, secretions, hardness, swelling, observed recorded. Photos taken area calculated on 1 st , 5 th 8 12 15 18 21 days. Hematoxylin-eosin (HE) staining utilized observe tissue 7 14 An enzyme-linked immunosorbent assay (ELISA) kit measure expression tumor necrosis factor (TNF)-α, interleukin (IL)-10, vascular endothelial growth (VEGF) transforming (TGF)- β 1. Results mainly include volatile oils, coumarins, lactones. UPLC–MS analysis revealed 39 main compounds extract. Among them, ferulic acid, kaempferitrin, caffeic salicylic acid have been confirmed exhibit anti-inflammatory antioxidant activity related burns scalds. HE gradual decrease number inflammatory cells healing wounds with increasing time extract administration. Compared group, showed significant differences levels TNF-α (161.67±4.93, 106.33±3.21, 77.67±4.04 pg/mL) IL-10 (291.77±4.93, 185.09±9.54, 141.33±1.53 days; difference content TGF- (75.68±3.06 day; level VEGF (266.67±4.73, 311.33±10.50 days respectively. Conclusion Petroleum oil might be drugs injuries, as they exhibited protective effect scalds reducing TNF-α, TGF-β1 VEGF. In addition, these may also exert pharmacological effects that promote repair, accelerate healing, reduce scar proliferation, inflammation pain.
