ABSTRACT
Extracellular bacteria, such as Pseudomonas aeruginosa and Klebsiella pneumoniae, have been reported to induce autophagy; however, the role machinery of infection-induced autophagy remain elusive. We show that pleiotropic Src kinase Lyn mediates phagocytosis autophagosome maturation in alveolar macrophages (AM), which facilitates eventual bacterial eradication. report is required for recruitment autophagic components pathogen-containing phagosomes. When we blocked with 3-methyladenine (3-MA) or by depleting Lyn, observed less subsequent clearance AM. Both morphological biological evidence demonstrated delivered bacteria lysosomes through xenophagy. TLR2 initiated phagocytic process activated following infection. Cytoskeletal trafficking proteins, Rab5 Rab7, critically facilitated early phagosome formation, maturation, autophagy-mediated degradation. These findings reveal Rab modulate related augment bactericidal activity, may offer insight into novel therapeutic strategies control lung
