ABSTRACT
The survival of central neurons depends on multiple neurotrophic factors produced by different cell types. We demonstrate that media conditioned muscle and Schwann lines show strong synergistic effects purified embryonic day 14.5 rat motoneurons in culture. Different evidence implicate glial line-derived factor (GDNF) cardiotrophin-1 (CT-1) this synergy. Their expression the environment motoneuron is compartmentalized: gdnf transcripts are expressed principally lines, whereas ct-1 mRNA present myotubes. Blocking antibodies to GDNF inhibit trophic activity line-conditioned 75%, CT-1 diminish myotube-derived 46%. act synergistically enhance vitro . In vivo , individual coexpress both receptor components. CT-1, therefore, major components support provided cells, respectively. possibility they together suggests must integrate distinct signals from cellular partners when deciding whether die or survive.
