ABSTRACT
Schwann cells are the main glial cell type in PNS. They develop along nerves during embryogenesis and rely on HMG domain containing Sox10 transcription factor for specification, lineage progression, terminal differentiation. deletion immature caused peripheral nerve defects mice that were not restricted to this type, although expression gene loss were. Formation of perineurium as protecting sheath was, instance, heavily compromised. This resembled defect observed after Desert hedgehog (Dhh) mice. Here we show activates Dhh via an enhancer is located intron 1 gene. binds monomeric form several sites. Mutation these sites abolishes both Schwann-cell-specific activity responsiveness vitro transgenic mouse embryos. argues by direct binding increasing levels promotes formation perineurial sheath. represents first mechanism a non-cell-autonomous function development.
