ABSTRACT
The effective use of MSCs for the treatment some B cell-mediated immune diseases is quite limited. main reason that immunomodulatory effects mesenchymal stem cells (MSCs) on are unclear, and their underlying mechanisms have not been fully explored.By co-culturing with without (MSC/CTLsh) or suppressor cytokine signaling 1 (SOCS1) knockdown (MSC/SOCS1sh), we found inhibited cell proliferation, activation terminal differentiation. Remarkably, highest inhibition proliferation was observed in MSC/SOCS1sh co-culture. Besides, reversed inhibitory effect last stage However, had no inhibiting by MSCs. We also showed IgA+ production significantly higher than MSC/CTLsh, although difference when both co-cultures were compared to isolated cells. In addition, increased PGE2 after TNF-α/IFN-γ stimulation, increase co-culture.Our results highlighted role SOCS1 as an important new mediator regulation function Therefore, these data may help develop treatments diseases.
