ABSTRACT
The transcription factor Pit-1/Pou1f1 regulates GH and prolactin (PRL) secretion in the pituitary gland. Pit-1 expression regulation by have also been demonstrated mammary However, no data are available on role of breast PRL. To evaluate this role, several human cancer cell lines were transfected with either vector or a small interference RNA construct, followed PRL mRNA protein evaluation. In addition, transient transfection MCF-7 cells reporter construct containing proximal promoter, ChIP assays performed. Our indicate that at transcriptional level binding to promoter. We found raises cyclin D1 before increasing levels, suggesting PRL-independent effect proliferation. By using immunohistochemistry, we significant correlation between 94 invasive ductal carcinomas.
