ABSTRACT
To ascertain the neuroendocrine function of kisspeptin/GPR54 system in non-mammalian species, full-length cDNAs encoding for Kiss1 and Kiss2 as well their putative cognate receptors GPR54a GPR54b, were isolated from goldfish ( Carassius auratus ). The deduced protein sequences between share very low similarity, but mature peptides (kisspeptin-10) are relatively conserved. RT-PCR analysis demonstrated that kiss1 gene gfkiss1 ) is highly expressed optic tectum-thalamus, intestine, kidney, testis, while kiss2 gfkiss2 mainly detected hypothalamus, telencephalon, tectum thalamus, adipose tissue, heart, gonads. two receptor genes gfgpr54a gfgpr54b brain regions including hypothalamus. Both kisspeptin-10 (gfKiss1–10 gfKiss2–10) biologically active they could functionally interact with cultured eukaryotic cells to trigger downstream signaling pathways different potencies. actions gfKiss1–10 gfKiss2–10 on LH secretion further investigated vitro vivo . Intraperitoneal administration sexually female increase serum levels. However, this peptide does not significantly influence release pituitary primary culture, indicating exert its at level. On other hand, appears be a much less potent it exhibits no significant bioactivity also inactive cells.
