ABSTRACT
In the central nervous system of mammals, there are specialized areas in which neurogenesis — neurogenic niches is observed postnatal period. It believed that astrocytes composition play a significant role regulation neurogenesis, and therefore they considered as promising “target” for possible control including use optogenetics. framework this work, we formed an vitro model niche, consisting cerebral endothelial cells, neurospheres. Astrocytes niche expressed canalorodopsin ChR2 underwent photoactivation. The effect photoactivated on expression profile cells was evaluated using immunocytochemical analysis methods. found intact contribute to neuronal differentiation stem well activation astroglia expressing photosensitive proteins, changes molecules characterized by intercellular interactions pools resting proliferating with participation NAD+ (Cx43, CD38, CD157), lactate (MCT1). particular, registered reflect violation paracrine two subpopulations one acts source NAD+, second consumer ensure processes intracellular signal transduction; change mechanisms transport due aberrant transporter MCT1 forming pool developing along path differentiation. general, photostimulation astrocytes, total proliferative activity increases mainly neural progenitor but not cells. Thus, optogenetic can become tool controlling formation local proneurogenic microenvironment niche.
