ABSTRACT
AIM: To explore the effects of hepatocyte growth factor (HGF) on retinal pigment epithelium (RPE) cell behaviors. METHODS: The human adult epithelial line-19 (ARPE-19) were treated by HGF or mesenchymal-epithelial transition (MET) inhibitor SU11274 in vitro. Cell viability was detected a Counting Kit-8 assay. proliferation and motility bromodeoxyuridine incorporation assay wound healing assay, respectively. expression levels MET, phosphorylated protein kinase B (AKT), AKT proteins determined Western blot MET also immunofluorescence RESULTS: increased ARPE-19 cells’ viability, migration, induced an increase proteins. significantly reduced proliferation, migration decreased suppressed HGF-induced cells. Additionally, blocked phosphorylation CONCLUSION: enhances cellular RPE cells through MET/AKT signaling pathway, whereas this enhancement is SU11274. might be vital contributor survival.
