Development of tryptophan metabolism patterns to predict prognosis and immunotherapeutic responses in hepatocellular carcinoma

ABSTRACT
Tryptophan metabolism is associated with tumorigenesis and tumor immune response in various cancers. Liver the main place where tryptophan catabolism performed. However, role of hepatocellular carcinoma (HCC) has not been well clarified. In present study, we described mutations 42 metabolism-related genes (TRPGs) HCC cohorts. Then, patients were distributed into two subtypes based on expression profiles TRPGs. The clinicopathological characteristics microenvironmental landscape profiled. We also established a TRPGs scoring system identified four hallmark TRPGs, including ACSL3, ADH1B, ALDH2, HADHA. Univariate multivariate Cox regression analysis revealed that TRPG signature was an independent prognostic indicator for patients. Besides, predictive accuracy assessed by receiver operating characteristic curve (ROC) analysis. These results showed risk model had excellent capability predicting survival both TCGA GEO Moreover, discovered significantly related to different infiltration therapeutic drug sensitivity. functional experiments immunohistochemistry staining validated above. Our comprehensive enhanced our understanding HCC. A novel built, which may be considered as beneficial tool clinical outcomes
