Prognostic and immunological roles of IL18RAP in human cancers

ABSTRACT
Across several cancers, IL18 receptor accessory protein (IL18RAP) is abnormally expressed, and this abnormality related to tumor immunity heterogeneous clinical outcomes. In study, based on bioinformatics analysis, we discovered that IL18RAP the human microenvironment promotes various immune cells infiltration. Additionally, multiple immunofluorescence staining revealed with increased expression of IL18RAP, number infiltrated M1 macrophages increased. This finding was confirmed by coculture migration analysis using three cancer cell lines (MDA-MB-231, U251, HepG2) knockdown. We a positive link between majority immunostimulators, immunoinhibitors, major histocompatibility complex (MHC) molecules, chemokines, chemokine genes Spearman correlation analysis. functional IL18RAP's gene set enrichment (GSEA) it variety immunological processes, such as regulation interferon gamma production NK cell-mediated immunity. Moreover, used single-cell RNA sequencing detect mainly expressed in cells, HALLMARK INF-γ upregulated CD8Tex cells. addition, mouse cohorts, found level can predict immunotherapy response. short, our study showed new biomarker may become potential immunotherapeutic target cancer.
