The inhibitory activity of gallic acid against DNA methylation: application of gallic acid on epigenetic therapy of human cancers

ABSTRACT
Epigenome aberrations have been observed in tobacco-associated human malignancies. (-)-epigallocatechin-3-gallate (EGCG) has proven to modulate gene expression by targeting DNA methyltransferases (DNMTs) through a proposed mechanism involving the gallate moiety of EGCG. We show that gallic acid (GA) changes methylome lung cancer and pre-malignant oral cell lines markedly reduces both nuclear cytoplasmic DNMT1 DNMT3B within 1 week. GA exhibits stronger cytotoxicity against line H1299 than found reactivates growth arrest damage-inducible 45 (GADD45) signaling pathway may demethylation CCNE2 CCNB1 cells. To improve epigenetic anti-cancer activities oolong tea, we identified fungus, Aspergillus sojae which can efficiently increase content tea via 2-week fermentation process. The fungus dramatically increased up 44.8 fold post-fermentation extract (PFOTE), resulting enhanced effects significant reduction abundances DNMT1, DNMT3A, lines. PFOTE also showed anti-proliferation (OTE) sensitivity cisplatin In summary, demonstrate potent inhibitory on DNMTs provide strong scientific foundation for use specialized fermented high as an effective dietary intervention strategy cancers.
