[Mechanism of Kuntai Capsules in treatment of polycystic ovary syndrome rat models based on AGE-RAGE signal pathway].

This study aims to investigate the impact of Kuntai Capsules(KTC) on polycystic ovarian syndrome(PCOS) rat models and explore underlying mechanism. Fifty female SD rats were randomly divided into five groups(10 in each group), including control group, model low-, medium-, high-dose KTC group. Except for other groups injected with dehydroepiandrosterone(DHEA) combined a high-fat diet(HFD) induce PCOS 28 days. 0.315, 0.63, 1.26 g·kg~(-1)·d~(-1) was dissolved same amount normal saline given by gavage. The pathological morphology tissue observed hematoxylin-eosin(HE) staining. expression levels collagen α type Ⅲ 1 chain(COL3A1), apoptotic factors Bax, Bcl-2 using Western blot immunofluorescence. mRNA expressions COL3A1, performed real-time PCR(RT-PCR). results show that compared body weight, serum FBG, FINS, LH, T, LH/FSH, HOMA-IR are higher group(P<0.05 or P<0.01), level FSH is lower(P<0.05). In large number white cells found vaginal exfoliated cells, mainly interictal phase. There more cystic prominences surface ovary. thickness granular cell layer reduced, oocytes absent. COL3A1 Bax protein increased(P<0.01), while decreased(P<0.05) increased tissue(P<0.05). Compared decreased(P<0.05 increased(P<0.05 P<0.01). Low-, gradually stable ovary smooth. Oocytes mature follicles can be seen tissue, increased. decreases low-and medium-dose groups(P<0.05 low-dose increases group(P<0.01). decreased group(P<0.05), increased(P<0.05). summary, inhibit granulosa apoptosis reduce follicular atresia regulating AGE-RAGE signaling pathway. It promote secretion, sugar restore hormone levels, improve symptoms PCOS, alleviate morphological damage ovary, function, which great value treatment PCOS.
