[Immune mechanism of Daphnes Cortex and its processed products on CIA rats based on B7/CD28/CTLA-4 pathway].

This study investigates the effects of Daphnes Cortex and its processed products on differentiation Th17/Treg cells in SD rats with type Ⅱ collagen-induced arthritis(CIA).Sixty-four were randomly divided into normal group(normal),model group(model),fried Daphne giraldii Nitsche low-dose high-dose groups(FDGN-L group, FDGN-H group),raw D. groups(RDGN-L RDGN-H group),daphnetin group(DAPH group),and tripterygium glycosides group(GTW group).Except for CIA model was immunized seventh day after first immunization, gavaged 28 days second immunization.After sampling, inflammation articular synovial membrane observed by hematoxylin-eosin(HE)staining; levels transforming growth factor-β(TGF-β),interferon-γ(IFN-γ),interleukin(IL)-2,IL-4,and IL-10 serum detected enzyme-linked immunosorbent assay(ELISA); real-time reverse transcription-PCR(qRT-PCR)and Western blot used to detect mRNA protein expressions cluster differentiation(CD) 80(B7-1),CD 86(B7-2),CD28,and cytotoxic T lymphocyte-associated antigen 4(CTLA-4)in rats; flow cytometry proportion Th17 Treg rats.The results showed that compared joint group significantly aggravated, arthritis index increased, immune organ increased(P<0.01).Compared each drug administration could improve varying degrees, reduce index, inhibit hyperplasia, index; IL-2 IFN-γ decreased(P<0.01),TGF-β,IL-4,and increased(P<0.01),the B7-1 CTLA-4 increased(P<0.01),and tissue decreased(P<0.01).In conclusion, inhibits expression promotes regulating B7/CD28/CTLA-4 pathway balance Th17/Treg, thereby treating rheumatoid arthritis.
