ABSTRACT
OBJECTIVE To investigate the effect of arsenic disulfide (AS2S2) combined with itraconazole on proliferation, apoptosis and hedgehog pathway diffuse large B-cell lymphoma (DLBCL) cells. METHODS The human DLBCL cell OCI-LY3 was treated different concentrations AS2S2 itraconazole. Cell proliferation inhibition detected by CCK-8, rate determined flow cytometry. expression levels BCL-2, BAX, SMO GLi1 were Western blot. RESULTS viability decreased significantly at 24, 48 or 72 h as cultured Along increasing concentration, reduced compared that in control group, results showed statistically significant(r=-0.690,r=-0.639, r=-0.833, r=-0.808, r=-0.578). inhibitory rates cells increased those single drug-treated group after combination AS2S2(P<0.05). protein Glil down-regulated alone(P<0.01). combined-treatment group(P<0.01). CONCLUSION Itraconazole can inhibit a concentration-and time-dependent manner. In addition, show synergistic effects, which may be related Hedgehog signaling pathway.
