ABSTRACT
The intracellular calcium concentration ([Ca2+]i) modulation plays a key role in the regulation of cellular growth and survival normal cells failure [Ca2+]i homeostasis is involved tumor initiation progression. Here we show that inhibition Furin by its naturally occurring inhibitor prodomain ppFurin MDA-MB-231 breast cancer resulted enhanced SOCE through TRPC6 activation associated reduced malignant phenotype. Expression stable manner melanoma MDA-MB-435 cell lines inhibits activity as assessed vitro digestion assays. Accordingly, transfection experiments, revealed ppFurin-expressing are unable to process adequately PC substrates proVEGF-C proIGF-1R. Compared cells, expression significantly induces Ca2+ entry which impaired silencing expression. Analysis up-regulated tyrosine phosphorylation cells. their viability ability migrate sensitization apoptosis inducer hydrogen peroxide. These findings suggest may be useful strategy interfere with mobilization leading phenotype repression reduction resistance treatments.
