ABSTRACT
Colorectal cancer (CRC) is a malignant tumor with high morbidity and mortality, but there still no recognized prognostic prediction model to better predict intervene its prognosis. Our aim establish novel microRNA (miRNA) signature identify hub target genes for simply accurately predicting survival risk CRC patients provide therapeutic targets. The miRNA expression profiles along clinical data of 512 were downloaded from the Cancer Genome Atlas (TCGA) database randomly divided into training set validation set. The was generated after series Cox regression analyses, including least absolute shrinkage selectionator operator (LASSO)-Cox regression, verified in test whole Furthermore, compared factors. Interaction network seven micoRNAs established. Functional enrichment analysis performed reveal biological processes pathways. GEPIA2 used analysis.A 7-micoRNA areas under receiver operating characteristic (ROC) curve (AUC) 5-year rate 0.889. Its performance well both entire by Kaplan-Meier (P value <0.05). Further demonstrated that an independent factor predictive ability superior age tumor-node-metastasis (TNM) stage. found two major gene modules, they might be involved activation PI3K-Akt-mTOR p53 signaling pathways, which related epidermal growth receptor (EGFR) resistance. revealed CDKN1A, eIF4E SNAI1 associated prognosis. Our study potential this independently overall provided targets.
