ABSTRACT
Objective – obtaining and characterization of hybrid cell lines producing monoclonal antibodies against I II types shiga-like toxins. Materials methods. Shiga-like toxins obtained in “48thCentral Research Institute” Ministry Defense Russian Federation (Kirov), BALB/c mice, myeloma cells SP2/0-Ag14 were used research. Immune splenocytes fused according to G. Kohler C. Milstein method De St. Fazekas D. Scheidegger modifcation using 50 % polyethylene glycol. Hybrid specifc cloned by limited dilutions. Hybridomas growth properties studied vitro vivo. Specifc activity immune sera, culture ascitic fluids indirect ELISA. Monoclonal from precipitated saturated ammonium sulfate, followed ion exchange chromatography Results discussion. 8 hybridomas obtained. are characterized stable proliferation antibody-producing during 10 passages 3 vivo (observation period). Obtained can be for ELISA detection Minimum detectable concentration sandwich is 1 ng/ml. The possibility detecting without typical differentiation was shown when the enzyme immunoassay a polyreceptor mixture sensitizing plate polyspecifc immunoperoxidase conjugates.
