ABSTRACT
Abstract Background Oral microbiome and salivary proteins play a critical role in the occurrence development of caries. In this study, we used metagenomic metaproteomic analyses to explore microbiological proteinic biomarkers investigate etiology caries preschool children. Our study aims offer better comprehension these factors relationship with caries, findings might facilitate risk assessment provide basis for future prevention strategies.Methods Children six eight years old living rural isolated areas or without were recruited. Supragingival plaque unstimulated saliva collected 16S rDNA pyrosequencing isobaric tags relative absolute quantitation (iTRAQ)technique coupled quantitative nano-flow liquid chromatography-tandem mass spectrometry(LC-MS/MS), respectively.Results A total 328486 high-quality rRNA sequences obtained from 40 samples including 20 subjects, which was clustered into 14,076 OTUs, representing 18 phyla, 28 classes, 48 orders, 78 families,135 genera, 410 species. We found most abundant phyla (Firmicutes, Bacteroidetes, Fusobacteria, Proteobacteria, Actinobacteria, Candidate division TM7). The Alpha diversity analysis demonstrated that richness bacterial communities similar between caries-free children; differences community composition analyzed by LEfSe. core defined as predominant genera coming subjects. For results proteome analysis, 9135 unique peptides 1662 group identified samples. 258 differentially expressed caries-active group, be potential biomarker health status.Conclusions Dental is multifactorial disease affecting many children around world. result our revealed complexities oral confirmed “ecological hypothesis”. addition, an important host factor are different healthy carious status.
