ABSTRACT
Abstract Background The cultivation and expansion of chicken primordial germ cells (cPGCs) is critical importance for both biotechnological applications the management poultry genetic biodiversity. feeder-free culture system has become most popular approach cells. However, despite some success in PGCs, reproducibility conditions across different laboratories remains a challenge. This study aimed to compare two defined enriched media growth cPGCs originating from Hubbard JA57 broiler. Methods To this end, were isolated embryonic blood 14–16 Hamburger–Hamilton (HH) stages cultured at various time points. Growth properties characteristics these evaluated their migratory assessed after engineering injection into vasculature 2.5-day-old embryos. Results main finding was that use an medium resulted improved PGCs compared medium. Conclusions ability cultivate expand crucial key approaches robust supportive can help accelerate generation genetically engineered birds avain transgenesis breeds interest local or commercial origins. Therefore, results highlight choosing appropriate cPGC expansion.
