ABSTRACT
New leaf rust resistance genes are needed in wheat ( Triticum aestivum L.) to provide additional sources of the highly variable and dynamic pathogen Puccinia triticina Eriks. Leaf gene Lr51 , located within a segment speltoides Taush chromosome 1S translocated long arm 1B bread wheat, is resistant current predominant races USA. The objectives this study were determine genetic length segment, develop PCR marker for use generate isogenic lines gene. Characterization two translocation (F‐7‐3 F‐7‐12) with 10 molecular markers indicated that F‐7‐3 has an interstitial T. 14 32 cM including loci XAga7 Xmwg710 whereas line F‐7‐12 complex series translocations among chromosomes homeologous group 1. On basis DNA sequence A, B, D, S alleles locus, we designed cleavage amplified polymorphic (CAPS) genome allele. Primers S30‐13L AGA7‐759R preferentially (819 bp) (783 bp). These amplification products can be separated agarose gels after digestion Pst I or Bam HI restriction enzymes. This CAPS was validated collection common cultivars used three pairs hard red spring from donor parent F‐7‐3. will valuable future assessment effect introgression on agronomic performance end‐use quality.
