ABSTRACT
Purpose: Arecoline is one of the main toxic components arecoline to cause oral mucosal lesions or canceration, which seriously affects survival and life quality patients. This study analyzed mechanism Jiawei Danxuan Koukang (JDK) in alleviating induced lesions, provide new insights for treatment submucosal fibrosis (OSF) cancerosis. Methods: Metabolomics was applied analyze composition JDK serum metabolites. The active ingredients were by combined ultra-high performance liquid chromatography mass spectrometry. target network JDK, metabolites OSF pharmacology, molecular docking. Oral HE Masson staining. Cell differentiation, proliferation apoptosis detected. expressions α-SMA, Collagen I, Vimentin, Snail, E-cadherin, AR NOTCH1 detected Western blot. Results: gradual atrophy thinning rat mucosal, collagen accumulation, increase fibrosis-related proteins Th17/Treg ratio. inhibited arecoline-induced inflammatory infiltration. changes Aminoacyl-tRNA biosynthesis, Alanine, aspartate glutamate metabolism Arginine biosynthesis pathways, reversed M-JDK. Quercetin key targets interaction. promoted expression protein, fibroblasts. effect on fibroblasts, but overexpression. CAL27 SCC-25 cells, cell proliferation, M-JDK quercetin. Conclusion: improved metabolite functional pathway. targeted protein improve OSF. quercetin cells play an anti-oral cancer role. Keywords: submucous fibrosis, Koukang, arecoline, metabolomics
