ABSTRACT
Yu Nu compound (YNJ) is a traditional Chinese medicine widely utilized to treat type 2 diabetes possibly through mediating autophagy. Abnormal podocyte autophagy and apoptosis could result in loss diabetics nephropathy (DN). The mechanism of DN still unclear. Therefore, the study aims investigate effects analyze potential mechanism.Goto-Kakizaki (GK) rats were administered using YNJ with different doses once day by gavage for 4 weeks. renal cortex injury was observed HE staining electron microscope. Cell analyzed TUNNEL staining. mTOR, autophagy-related proteins apoptosis-related detected Western blot or real-time PCR vivo vitro. MPC5 cells exposed high glucose (HG, 30mM) 12h simulate DN. treated serum containing dosages. activities were, respectively, Counting Kit-8 (CCK8) assay flow cytometry.The results showed that medium dose had better on decreasing blood improving GK rats, followed mTOR levels. levels enhanced cortex, accompanied increase cell vivo. Besides, regulating significantly modulated rats. Then, we found endogenous reverse vivo.The suggested recovered normal suppressed mTOR. maintenance basal autophagic activity based effect multiple target essential maintaining function.
