ABSTRACT
Purpose: The primary goal of the present study was to develop nano-drug consisting doxorubicin and exosome derived from mesenchymal stem cells, explore its effect on osteosarcoma in vitro. Methods: exosomes were isolated bone marrow MSCs (BM-MSCs) by an Exosome Isolation Kit. exosome-loaded (Exo-Dox) prepared mixing with Dox-HCl, desalinizing triethylamine then dialyzing against PBS overnight. nanoparticle tracking analysis (NTA) transmission electron microscope (TEM) used characterize Exo-Dox. cytotoxicity Exo-Dox determined CCK-8 assay. Further, cellular uptake different drugs analyzed using inverted fluorescence flow cytometry. Results: typical structures can be observed TEM. After loading doxorubicin, size is larger than free exosome. Compared Dox, showed enhanced efficiency anti-tumor MG63 cell line but low myocardial H9C2 line. Conclusion: could as excellent chemotherapeutic drug for treatment Considering tumor-homing feature BM-MSCs, may a good candidate targeted future study. Keywords: exosome,
