ABSTRACT
Purpose: The first-line chemotherapy treatment protocol for gastric cancer is combination of 5-fluorouracil (5-FU) and cisplatin (CDDP). aim this study was to engineer prodrug-based nanostructured lipid carriers (NLC) platform codelivery 5-FU CDDP enhance therapy decrease toxicity. Methods: First, 5-FU-stearic acid conjugate synthesized by two steps. Second, prodrug were loaded in NLC. Finally, hyaluronic (HA) coated onto NLC surface. Average size, zeta potential, drug loading capacity evaluated. Human cell line BGC823 (BGC823 cells) used the testing vitro cytotoxicity assays. In vivo antitumor activity evaluated mice bearing cells model. Results: HA-coated CDDP-loaded (HA-FU/C-NLC) showed a synergistic effect displayed greatest than all free drugs or uncoated vivo. Conclusion: This work reveals that could be as novel carrier codeliver therapy. HA-FU/C-NLC promising targeted combinational nanomedicine. Keywords: cancer, carriers, acid, chemotherapy,
