ABSTRACT
This study aims to screen microRNAs (miRNAs), for an early diagnosis of colorectal cancer, by deep sequencing and evaluation total miRNAs using clinical samples from a Chinese patient population.Total small RNAs normal colonic mucosa, adenomas, cancer tissues were prepared miRNA analysis sequencing. The data then analyzed bioinformatics candidate diagnostic miRNAs, which further validated their up- or downregulation status.Comparison with mucosa identified 99 upregulated 90 downregulated miRNAs. Comparison adenomas found 114 107 adenoma 70 27 Selected expressions in 12 cases patients polyps. Specifically, the miR-18a-5p miR-21-3p significantly tissues, compared mucosa; miR-135b-5p, miR-17-5p, miR-182-5p, miR-200a-5p, miR-200c-3p but differential expression was not significant when mucosa. miR-183-5p miR-96-5p these differences For miR-133a-3p both miR-204-5p, miR-125b-5p, miR-139-5p, miR-100-5p, miR-30a-5p tissue mucosa. The findings this show that number might be important prognosis method RNA Upregulation may serve as index screening cancer. Other such miR-200c-3p, miR-183-5p, miR-96-5p, either downregulated, have limited significance diagnosis. Further is needed determine value.
